Nilonix Ingredients and Usage

Nilonix 200mg Capsule contains nilotinib hydrochloride monohydrate, a second generation tyrosine kinase inhibitor designed to block the BCR ABL fusion protein. Beacon Pharmaceuticals manufactures the capsule in a hard gelatin shell for oral administration twice daily. Each capsule delivers 200mg of nilotinib, the standard unit dose used in adult oncology regimens.

Doctors prescribe Nilonix for adults with Philadelphia chromosome positive chronic myeloid leukemia in chronic and accelerated phase. The drug suits both newly diagnosed patients and those resistant or intolerant to imatinib therapy. Nilonix produces measurable hematologic, cytogenetic, and molecular responses in CML, restoring normal blood cell production over weeks of dosing.

How Does Nilonix Work?

Nilonix works by binding to the ATP binding site of the BCR ABL fusion protein, blocking its tyrosine kinase activity. This abnormal protein arises from the Philadelphia chromosome translocation and drives uncontrolled white blood cell proliferation in CML. By inhibiting BCR ABL signalling, nilotinib stops leukemic cells from multiplying and triggers their natural cell death pathway.

Nilonix also inhibits related kinases including PDGFR and KIT, broadening its activity against resistant leukemia cell lines. Clinical studies show nilotinib delivers major molecular response rates of around 44 percent at 12 months in newly diagnosed CML. Patients take Nilonix twice daily, 12 hours apart, on an empty stomach to ensure consistent absorption and predictable plasma levels.

Nilonix Side Effects

Nilonix can cause cardiac, gastrointestinal, and hematologic side effects, with QT interval prolongation being the most serious risk. Most reactions appear within the first months of therapy and require regular ECG and blood count monitoring.

Warnings

  • Nilonix carries a boxed warning for QT interval prolongation, which can cause sudden cardiac death.

  • Avoid Nilonix in patients with hypokalaemia, hypomagnesaemia, or long QT syndrome until electrolyte levels are corrected.

  • Strong CYP3A4 inhibitors such as ketoconazole, clarithromycin, and ritonavir raise nilotinib levels and the risk of cardiac toxicity.

  • Avoid grapefruit, grapefruit juice, and proton pump inhibitors, as these alter nilotinib absorption and reduce efficacy.

  • Take Nilonix on an empty stomach with no food 2 hours before and 1 hour after each dose.

  • Hepatic impairment requires dose reduction, and liver function tests must be monitored throughout treatment.

  • Pregnant or breastfeeding women must not use Nilonix due to documented risk of fetal harm and infant toxicity.

  • Inform your doctor about any history of heart disease, recent heart attack, or stroke before starting Nilonix.

Nilonix Storage Conditions
