Regonix Ingredients and Usage
Regonix 40mg Tablet contains regorafenib, a multi kinase inhibitor that targets several enzymes driving tumour growth and angiogenesis. Beacon Pharmaceuticals manufactures the tablet as a light pink, oval, film coated dose for oral once daily administration. Each tablet delivers 40mg of regorafenib, with a typical treatment dose of 160mg given as four tablets daily.
Doctors prescribe Regonix for adults with metastatic colorectal cancer who have failed fluoropyrimidine, oxaliplatin, irinotecan, anti VEGF, and anti EGFR therapy. The medicine also treats unresectable gastrointestinal stromal tumours after imatinib and sunitinib failure, and hepatocellular carcinoma after sorafenib. Regonix offers a salvage option that prolongs progression free survival in patients with limited remaining treatment choices.
How Does Regonix Work?
Regonix works by blocking multiple receptor tyrosine kinases on tumour cells and the surrounding blood vessels. The drug inhibits VEGFR1, VEGFR2, and VEGFR3, cutting off the new blood vessel formation that solid tumours need to grow. Regonix also targets PDGFR, FGFR, KIT, RET, and the RAF kinases involved in cancer cell survival and proliferation.
This broad spectrum kinase inhibition starves tumours of blood supply, halts cell division signals, and triggers programmed cell death. Clinical trials show Regonix improves median overall survival by around 1.4 months in metastatic colorectal cancer versus placebo. Patients take Regonix once daily for the first 21 days of each 28 day treatment cycle, with a low fat meal.
Regonix Side Effects
Regonix can cause skin, gastrointestinal, and liver side effects, with hand foot skin reaction and hepatotoxicity being the most prominent. Most reactions appear within the first two months of treatment and need close monitoring through regular blood tests.

Warnings
Regonix carries a boxed warning for severe and sometimes fatal liver injury, with most cases occurring within the first two months of treatment.
Hyperammonemic encephalopathy has occurred in patients with liver dysfunction, liver metastases, or primary liver cancer taking regorafenib.
Regonix increases the risk of serious bleeding and gastrointestinal perforation, which may require permanent discontinuation.
Hand foot skin reaction affects more than half of patients and may require dose reduction or temporary treatment interruption.
Withhold Regonix for at least two weeks before elective surgery and resume only after complete wound healing.
Strong CYP3A4 inhibitors such as ketoconazole and clarithromycin raise regorafenib levels and increase the risk of toxicity.
CYP3A4 inducers like rifampin, phenytoin, and St. John's Wort lower drug exposure and reduce treatment efficacy.
Avoid grapefruit and grapefruit juice during Regonix treatment due to interference with drug metabolism.
Pregnant women must not use Regonix due to risk of embryo fetal harm, and breastfeeding is not recommended.
Monitor blood pressure regularly, as Regonix commonly causes new or worsening hypertension during treatment.
